摘要
目的:探讨葛根素对实验性糖尿病大鼠胰腺线粒体自由基损伤的保护作用。方法:30只Wistar大鼠随机分为正常对照组、糖尿病组和治疗组。采用四氧嘧啶腹腔注射复制糖尿病动物模型。葛根素注射液治疗8周后,测定血清葡萄糖、胰岛素、胰腺线粒体丙二醛(MDA)含量及超氧化物歧化酶(SOD)、Na+-K+-ATP酶(Na+-K+-ATPase)和Ca2+-ATP酶(Ca2+-ATPase)的活性。结果:糖尿病组与对照组比较,胰腺线粒体膜出现脂质过氧化,Na+-K+-ATPase和Ca2+-AT-Pase活性下降。使用葛根素治疗后,胰腺各项测定指标明显改善。结论:葛根素对糖尿病大鼠胰腺线粒体自由基损伤有明显的保护作用。
Objective:To investigate protective effect of of puerarin on pancreatic mitochondria injury caused by free radicals in diabetic rats.Methods:30 rats were divided into control group,diabetes group and treatment group at random.Diabetic model was reproduced by intraperitoneal injection of alloxan.After having been treated with puerarin for 8 weeks,we measured the serum contents of glucose 、insulin 、MDA,the activity of superoxide dismutase(SOD),Na+-K+-ATPase and Ca2+-ATPase in pancreatic mitochondria were detected respectively by kit.Results:Compared with control group,pancreatic mitochondria showed lipid peroxidase injury,the activity of Na+-K+-ATPase and Ca2+-ATPase was reduced.All the detection indexes in pancreatic were significantly improved after therapy with puerarin.Conclusion:Puerarin might have protective effects on the pancreatic mitochondria injury caused by free radicals in diabetic rats.
