摘要
[目的]建立甜瓜再生体系。[方法]以加格达甜瓜为材料,以MS培养基为基本培养基,根据植物生长调节剂对植物愈伤组织的诱导分化的决定性作用,通过设计不同生长素和细胞分裂素的组合与不同浓度配比,研究建立和优化高效的加格达甜瓜再生体系。[结果]愈伤组织诱导培养基:MS+2,4-D 1.5 mg/L+6-BA 0.1 mg/L,出愈率93%;芽诱导培养基:MS+6-BA 2.0 mg/L,出愈率76%,出芽率100%;芽的分化培养基:MS+6-BA 2.0 mg/L+IAA0.2 mg/L,分化率为95%;芽的伸长培养基为:MS+6-BAI 0.05 mg/L;根的诱导培养基为:MS+IAA0.2 mg/L。[结论]建立了1套甜瓜的离体再生体系,为利用转基因技术培育出具有抗病虫害、抗逆性等优良性状的甜瓜奠定基础,在理论和实践上均具有重要的意义。
[Objective] The research aimed to establishing the regeneration system of Cucumis melo L. vat. sacchanus Naud.. [Method] Jiageda melon variety was used as research material and MS as basic medium. For the dominant function of hormone to the plant growth, different auxin and cytokinins were selected and different concentration was used on Cucumis melo L. var. sacchanus Naud, tissue culture, the high-efficient regeneration system of Cucumis melo L. vat. sacchanus Naud. was studied. [Result] The callus induction medium was MS+2,4-D 1.5 mg/L+6-BA 0.1 mg/L, and the callus induction rate reached 93%. The shoot induction medium was MS+6-BA 2.0 mg/L, and the shoot induction rate reached 76%. The shoot differentiation medium was MS+6-BA 2.0 mg/L+IAA 0.2 mg/L, and the differentiation rate reached 95%. The shoot elongation medium was MS+6-BAI 0.05mg/L. The root induction medium was MS+IAA 0.2 mg/L. [Conclusion] It was important to establish a set of technique for the regeneration of Cucumis melo L. var. sacchanus Naud., which laid the foundation for cultivating the Cucumis melo L. vat. sacchanus Naud. varitey with disease and inset pests resistance and stress resistance by using transgenic technology, and had important signification in theory and practice.
