摘要
对石榴果皮褐变产物进行了提取、分离和定性定量分析。探讨了超声波法提取蒽醌类化合物的工艺条件,通过正交实验研究了提取温度、溶剂浓度、提取时间、溶剂体积四因素对蒽醌类化合物提取率的影响。结果表明,最佳提取工艺条件为:乙醇为提取溶剂,乙醇浓度75%,乙醇用量90mL,提取时间25min,提取温度25℃。并将超声波提取与溶剂回流提取进行了比较,得出超声波法优于传统的溶剂回流法。褐变产物鉴定为蒽醌类化合物,利用pH梯度萃取法将提取物分离出四种物质,且通过薄层层析鉴定出它们的结构。
The extraction, separation and qualitative appraisal on the browning product of the pomegranate peel were studied. The extraction technical conditions of the anthraquinones were discussed by the ultrasonic-assisted method. The influence of temperature, solvent concentration, time and solvent volume on the distilling rate of anthraquinones was studied through the orthogonal test. The result indicated that the best extraction technical conditions were that ethanol concentration 75%, ethanol volume 90mL, extraction time 25min, extraction temperature 25℃. By comparing the ultrasonic-assisted extraction with the solvent circumfluence extraction, we draw a conclusion that the ultrasonic-assisted was obviously superior to the traditional solvent circumfluence. The products were anthraquinones through qualitative appraisal, and four kinds of product were separated by the pH gradient solvent extraction method, each structure was identified by thin-layer chromatography.
