硫酸镁对兔视网膜缺血再灌注损伤的保护作用 被引量:1

摘要 目的探讨硫酸镁(MgSO4)对实验性兔视网膜缺血再灌注损伤(RIRI)的保护作用及可能的作用机制。方法眼内灌注法建立RIRI模型。32只白兔随机分为模型组、正常对照组、MgSO4治疗A组、MgSO4治疗B组。A组缺血后立即按140mg/kg耳缘静脉推注25%MgSO4,然后加用1g腹腔内注射;B组缺血后按600mg/kg腹腔内注射25%MgSO4。再灌注6h后采集视网膜组织,用硫代巴比妥分光光度法(TBA)测定丙二醛(MDA)含量,黄嘌呤氧化酶法测定超氧化物歧化酶(SOD)活性。结果再灌注6h后,各治疗组视网膜组织中SOD活性较模型组升高,MDA含量较模型组降低,并与给药途径有关。结论MgSO4可增加实验性RIRI的家兔视网膜组织中的SOD活性,降低MDA的含量,从而起到保护视网膜的作用。 Objective The ischemic retina diseases always result in visual dysfunction. The aim of this study was to investigate the protective effects of MgSO4 on high intraocular pressure (IOP)-induced retinal ischemia-reperfusion injury. Methods The model of experimental retinal ischemia-reperfusion injury was created by increasing IOP to 120 mmHg through intrachamber infusion of normal physiology salt solution for 45 minutes and then recoverred the lOP by lowing the highty of container in 32 white rabbits. The animal models were randomly divided into model group, normal control group and MgSO4 treatment group,and the latter was subdivided into the MgSO4 A and MgSO4 B group. 25% MgSO4 ( 140 mg/kg) was given by intravenous injection,and then 1 g of MgSO4 was injected intraperitoneally in MgSO4 treatment group A, and 600 mg/kg MgSO4 was given by intraperitoneal injection in MgSO4 treatment group B. The rabbits were sacrificed at 6 hours after reperfusion. The content of malondialdehyde(MDA) and the activity of superoxide dismutase(SOD) in retina was detected using spectrophotometer. Results The activity of SOD in retina in various groups showed a statistically significant difference ( F = 83.94, P 〈 0.05 ). Compared with normal control group,the content of retinal SOD was obviously elevated in model group,MgSO4 A and MgSO4 B groups(P 〈0.05). The content of MDA presented a obviously difference(F = 135.95 ,P 〈0. 05) and showed a gradually reduced tendency in model group,MgSO4 A and MgSO4 B groups in comparison with normal control group(P 〈 0.05). Conclusion MgSO4 has the protective effects on experimental retinal ischemia-reperfusion injury in rabbits by increasing the activity of SOD and decreasing the content of MDA.
