Differentiation of human embryonic stem cells into insulin-secreting cells

摘要 人类胚胎干(hES)细胞具有极其强大的增殖能力,理论上具有分化为机体所有组织细胞的潜能。治疗性克隆技术可用于制备带有患者基因型的hES细胞及其分化而来的胰岛素分泌细胞,但存在激烈的伦理学争论。新近,通过体外转基因处理可诱导人类体细胞重构形成hES样的多潜能干细胞。本文着重介绍hES细胞定向分化为胰岛素分泌细胞的研究进展。 Human embryonic stem (hES) cells exhibit remarkable long-term proliferative potential, and theoretically have the ability to form any fully differentiated cells of the body. Therapeutic cloning strategy can be used to generate hES cells and insulin-secreting cells with patients' genotype,but has raised intense ethical debate. It has been shown recently that reprogramming of human somatic cells to a pluripotent hEScell-like state can be induced in vitro by retroviral introduction of four transcription factors. This review discusses the advance in the differentiation of hES cells into insulin-secreting cells.
