摘要
目的观察脑梗死后亚低温干预对大鼠血管内皮生长因子(vascular endothelial growth factor,VEGF)和基质金属蛋白酶抑制剂-1(tissue inhibitor of metalloproteinase-1,TIMP-1)的表达变化,探讨亚低温对缺血后血脑屏障通透性的影响。方法线栓法制造大鼠脑缺血再灌注模型,同时给与亚低温治疗。用免疫组织化学方法检测VEGF、TIMP-1的表达。结果再灌注损伤后脑内VEGF和TIMP-1蛋白的水平开始升高,至再灌注后72h达最高水平,亚低温组VEGF的表达明显低于常温组,而TIMP-1明显高于常温组。结论亚低温能有效抑制VEGF的表达和增加TIMP-1的表达,减小血脑屏障的通透性,减轻脑缺血再灌注后脑水肿程度。
Objective To study the effects of mild hypothermia on VEGF(vascular endothelial growth factor,VEGF) and TIMP-1(tissue inhibitor of metalloproteinase-1,TIMP-1) expression and blood-brain barrier permeability in rat focal infarcted cerebral tissues,and investigate the mechanisms of mild hypothermia on ischemia brain edema.Methods The focal cerebral ischemia-reperfusion SD rat models were made by thread embolism in middle cerebral artery.Some of these rats were treated with mild hypothermia.The expression of VEGF and TIMP-1 were observed by immunohistochemical technique.Results The levels of VEGF and TIMP-1 expressions in the rats increased after focal cerebral ischemia and reperfusion.The highest point was at 72h after reperfusion.In the mild hypothermia group,the VEGF expression levels at every time point were obviously lower than those in the normal temperature group;but TIMP-1 levels were higher.Conclusion Mild hypothermia may relieve cerebral edema after cerebral infarct since it can inhibit VEGF and increase TIMP-1 expressions.
