摘要
背景与目的:血管性血友病因子(von Willebr and factor,vWF)、整合素β3(integrinβ3)与肿瘤转移的机制目前研究不清楚。本研究目的是观察vWF与integrinβ3在人肺癌细胞H460中的表达,人肺癌细胞H460与vWF黏附的关系,两因子与肿瘤轻移的相关性及vWF与integrinβ3的相互关系。方法:①应用免疫组化技术观察vWF及integrinβ3在人肺癌细胞株H460的表达;②联合应用vWF黏附实验、抗体抑制实验及MTT法,观察vWF及integrinβ3与人肺癌细胞H460的黏附,及vWF与integrinβ3的相互关系。结果:①免疫组化结果显示H460细胞均表达vWF及integrinβ3。②vWF可以与H460细胞黏附,Anti-integrinβ3抑制后这种黏附降低,A值由1.59±0.06降到0.55±0.03,(P=0.01619),Anti-vWF抑制后这种黏附也降低,A值由1.60±0.06降到0.54±0.03,(P=0.01598),两者抑制黏附效果相同。结论:H460细胞表达vWF及integrinβ3,vWF可以与H460细胞黏附,vWF可能通过与integrinβ3结合发挥黏附作用,从而促进肿瘤细胞转移。
Background and purpose: The impact of the two factors of von Willnbrang factor(vWF) and integrinβ3 on tumor metastasis has not been recognized. This study was done to investigate the expression of vWF and integrinβ3 in human lung cancer cell line H460, the association between the two factors and tumor metastasis effect of vWF and integrinβ3 on adhesion of human lung cancer cell line H460. Methods: The expression of vWF and integrin β3 protein was examined by immunohistochemical staining. The impact of vWF and integrinβ3 on adherent effect of tumor cells was evaluated by adhesion experiment, antibody inhibiting experiment and MTT. Results: Positive expression of vWF and integrinβ3 was detected in H460 human lung cancer cells. H460 human lung cancer cells were able to adhere to vWF. Using anti-integrinβ3, we observed that the ability of cell adhesion to vWF was inhibited, and A Value decreased from 1.59±0.06 to 0.55±0.03 ( P =0. 01619). Using anti-vWF, we observed cell adhesion to vWF was inhibited too and A Value decreased from 1.60±0.06 to 0.54±0.03( P = 0. 01598), which had the same effect when using anti-integrinβ3. Conclusions: H460 human lung cancer cells are capable of producing vWF, and vWF expression contributes to metastasis by adhering to cancer cells, integrinβ3 is the vWF receptor on H460 human lung cancer cells.
