摘要
四倍体龙牙百合鳞片叶直接形成不定芽和体细胞胚的培养及不定芽直接发生的生理生化特性测定结果表明:在MS培养基中添加一定浓度的6-BA可以诱导外植体直接分化不定芽。生长素2,4-D等则强烈抑制不定芽的直接分化。龙牙百合体胚发生的重要调节因素是2,4-D,体胚发生与2,4-D浓度及2,4-D的预培养时间密切相关。在不定芽直接分化的不同时期,蛋白质、核酸与ATP的含量及蛋白酶、POD、SOD的活性存在明显差异,各自又呈现一定的变化规律。上述物质的变化特性说明形态发生与建成受基因选择性表达的调控。
The direct formation of indefinite buds and somatic embryoes from scale leaf and the qualities of physiology and biochemistry of the period of bud formation were investigated. The results indicated that the indefinite buds were induced directly from the scale leaf by culturing them on MS medium containing cytokinin 6-BA, but Growth hormone, such as 2,4-D restrained bud direct differentiation. The somatic embryogenesis had closed relation to 2,4-D. The contents of protein, nucleic acid, ATP and the activities of protease POD and SOD of explants at different culture time showed significant difference.
