Effects of Basic Fibroblast Growth Factor on the Expression of Protoonco gene c fos and c myc in Rat Glomerular Mesangial Cells

摘要 在建立大鼠肾小球系膜细胞(MC)体外培养方法的基础上,通过3H-TdR参入实验,RNA印迹分析和斑点杂交观察bFGF对MCDNA合成及原癌基因c-fos和c-myc表达的影响.结果表明,bFGF作用于MC18h,MC的3H-TdR参入率明显增加(P<0.05),24h达到高峰(P<0.01);bFGF显著诱导原癌基因c-fos和c-myc表达,其表达活性分别于30min和1h达到高峰.提示bFGF是MC的强效丝裂原,其对MCDNA合成的促进作用与诱导原癌基因c-fos和c-myc表达有关. The culture method for rat glomerular mesangial cells (MCs) in vitro was established.The effects of basic fibroblast growth factor (bFGF) on DNA synthesis of MC and expression of protooncogene c fos and c myc in MC were determined by 3H TdR incorporation,Dot blot and Northern blot analysis.The results showed that the value of 3H TdR incorporated into MC significantly increased when MCs were incubated with bFGF(20 ng/ml) for 18 hours ( P< 0 05),and reached peak at 24 hours;bFGF significantly induced the expression of protooncogene c fos c myc.The maximal expression of protooncogene c fos and c myc was respectively at 30 min and 1 hour after induced by bFGF.These data indicate that bFGF is a stronger mitogen for MC,the stimulation of bFGF for DNA synthesis of MC relates to the expression of protooncogene c fos and c myc induced by bFGF.
