摘要
目的:探讨NOR1基因对HepG2细胞E-选择素的影响。方法:将NOR1基因通过脂质体转染HepG2细胞后,抽提细胞总RNA,采用RT-PCR法检测HepG2细胞中NOR1基因和E-选择素mRNA的表达水平;ELISA法检测细胞培养上清液中E-选择素的蛋白表达水平。结果:RT-PCR结果显示,转染NOR1基因组E-选择素mRNA的表达水平明显高于空白组和对照组的E-选择素mRNA的表达水平(P<0.01)。ELISA结果显示,转染NOR1基因组E-选择素蛋白水平明显高于空白组和对照组(P<0.01)。结论:NOR1基因可诱导HepG2细胞中E-选择素表达水平的增高。
Objective:To study the effect of NOR1 gene on expression of E-selectin in HepG2 cells. Methods:NOR1 gene was transfected into human liver cancer HepG2 cells mediated by cationic lipofectamine. The total RNA was extracted. The mRNA expressions of NOR1 gene and E-selectin in HepG2 cells were detected by reverse transcription polymerase chain reaction (RT-PCR). The level of E-selectin protein in the supematant of cell culture was determined by ELISA method. Results:The results of RT-PCR showed that the mRNA expression of E-selectin in NOR1 gene transfection group was significantly higher than those in blank group and control group (P 〈0.01 ). The results of ELISA showed that the protein level of E-selectin in the supematant of cell culture was significantly higher in NOR1 gene transfection group than those in the blank group and control group (P 〈 0.01 ). Conclusion:NOR1 gene could induce the increase in E-selectin expression in HepG2 cells.
