摘要
目的通过放射线60Co照射SD大鼠,观察大鼠视网膜毛细血管内皮细胞的改变,以探讨放射性视网膜病变发病机制。方法45只雄性SD大鼠随机分成正常对照组、10Gy组、30Gy组(每组15只),60Co照射大鼠制备实验模型。之后分别于照射之后1个月、3个月、6个月处死10Gy、30Gy、正常对照组各1组动物(每组5只)。做大鼠视网膜铺片,光镜观察毛细血管内皮细胞数量。结果正常对照组大鼠随时间的延长,视网膜毛细血管内皮细胞数量无显著差异(P>0.05)。而模型组与对照组比较:检测毛细血管内皮细胞数量:①1个月,10Gy、30Gy与对照组比较无显著差异;②3个月:30Gy组与对照组有显著差异(P<0.05);③6个月:30Gy组与对照组有极显著差异(P<0.01);④对照组间随时间改变比较无显著差异;⑤6个月:10Gy组与对照组比较有显著差异(P<0.05)。结论放射线60Co照射SD大鼠后,大鼠视网膜毛细血管受损,毛细血管内皮细胞数量减少,并随着实验时间延长和剂量的增加愈加严重。这就为探讨放射性视网膜病变发病机制提供实验依据。
Objective We use radioactive ray to irradiate SD rats, Observing the detriment of the radioactive ray to rat's retina blood capillary, investigating the mechanism of the radiation retinopathy onset. Methods 45 male Spregue - Dawley(SD) rats model were made by ^60Co dioative - ray and randomly divided into normal control group, 10Gy group and 30Gy group of dioative - ray rats. Each group has 15 rats. Putting the rats to death of every groups (In every 1,3,6months): 10Gy,30Gy, normal ones (each group: 5 ). Making the stretched preparation of the rat' s retina. Observing the numbers of endothelial cells with the light microscope. Results With extend of course of disease of normal control, the numbers of endothelial cells have no significant deviation (P 〉 0.05 ). Comparison group model and normal control: detected the numbers of endothelial cells: (1)1 month: control group contrast with test group has no changed;(2)3 month: group 30Gy/N ( P 〈 0.05 ) ; (3)6 month : Group 30Gy /N ( P 〈 0.01 ) ; (4)With time pass, control group has no changed; (5) Group10Gy :Group 6 month/N(P 〈 0.05). Conclusion After rats have be radiated by radioactive ray 60Co, their retina blood capillary has been damaged, the numbers of endothelial cells have been decreased. With extended experimental session and increased dose, symptom has graveness. That will offer theory for approach nosogenesis of radiation retinopathy.
