RNA干扰技术在逆转肿瘤多药耐药中的应用研究

摘要 RNA干扰(RNAi)指与内源性mRNA编码区某段序列同源的双链RNA导入细胞时,该序列mRNA发生特异性的降解,并导致基因表达的沉默。小干扰RNA(siRNA)是RNAi作用的主要介质,其可经体外合成再转入细胞,也可通过各种载体内源性表达。RNAi为肿瘤的基因治疗提供新的思路,通过RNAi抑制肿瘤相关耐药基因的表达有望成为一种逆转肿瘤耐药新的策略。 RNA interference (RNAi) is the process of sequence specific pose-transcriptional gene silencing intiated by homologous double-stranded RNA. Small interfering RNA (SiRNA) is the main medium in the process of RNAi, which can be syntheticed in vitro and then transfected into cell or endogenously expressed by vectors. RNAi provides a new thought for gene-therapy of tumor. Inhibition of drug resistance-associated gene expression through RNAi is hopeful to be a new strategy for reversal drug resistance in tumor.
