前列腺癌中血管生成素-2及其受体Tie2的表达及与血管生成的关系

Expressions of Ang-2 and its receptor Tie2 in squamous cell carcinoma of the prostate and their correlation with angiogenesis

摘要 目的:探讨血管生成素-2(Ang-2)及其受体Tie2在前列腺癌组织中的表达规律及其在肿瘤组织发生、发展以及血管生成中的作用.方法:采用免疫组织化学S-P染色法检测39例前列腺癌组织中Ang-2,Tie2及CD34的蛋白表达,根据CD34染色结果计数微血管密度(MVD).结果:Ang-2,Tie2的蛋白表达定位于肿瘤细胞或血管内皮细胞质及胞膜,呈异质性分布;在39例前列腺癌中Ang-2和Tie2阳性表达率均为74%;且均与前列腺癌的分化程度、淋巴结转移密切的关系(P<0.05);Ang-2,Tie2蛋白阳性表达者的MVD分别为(42.1±5.2)和(42.3±5.1),显著高于相应的前列腺癌组织Ang-2和Tie2阴性病例[MVD分别为(33.7±3.3)和(33.2±2.3)](P<0.01).结论:Ang-2,Tie2在前列腺癌发生、演进过程中起着重要作用,并与肿瘤血管生成关系密切. AIM: To investigate the expression patterns of angiopoietin-2(Ang-2) and its receptor, tyrosine kinase with immunoglobulin and epidermal growth factor homology domains (Tie2) in the squamous cell carcinoma of the prostate and evaluate their roles in the tumor initiation, development and angiogenesis. METHODS: Expressions of Ang-2, Tie2 and CD34 proteins were detected immunohis to chemically in 39 cases of the differently graded squamous cell carcinoma of prostate. Microvessel density(MVD) was counted according to the result of CD34 protein staining. RESULTS: Ang-2 and Tie2 were mainly localized in the cytoplasm and plasmalemma of tumor cells and endothelial cells in an obvious heterogeneity fashion. The positive rates of both Ang-2 and Tie2 proteins were 74%. The levels of the two proteins were significantly related to differentiation degree and lymphatic metastasis of tumor cells( P 〈 0. 05 ). The MVDs of the cases of Ang-2 and Tie2 strongly-positive staining were (42. 1 ± 5.2) and (42.3 ± 5.1 ), respectively, both of which were significantly higher than those of negative staining (33.7 ± 3.3, 33.2 ± 2.3, P 〈0.01 ). CONCLUSION: Both Ang-2 and Tie2 proteins might be of much importance in the carcinogenesis and progression of squamous cell carcinoma of the prostate. They might also play pivotal roles in neoangiogenesis.
