摘要
目的:观察实验性肾阳虚大鼠生殖细胞凋亡及Bcl-xl、Bad蛋白表达的变化。方法:12只成年健康雄性SD大鼠随机分成肾阳虚组(n=6)和对照组(n=6)。建立单侧肾阳虚动物模型后,取睾丸,对照组同侧取材。采用原位缺口末端标记法(TUNEL)检测生殖细胞凋亡,免疫组化法检测Bcl-xl、Bad蛋白表达。结果:凋亡细胞主要是初级精母细胞和圆形精子细胞。肾阳虚组生殖细胞凋亡指数高于对照组(P<0.05);肾阳虚组Bcl-xl蛋白表达低于对照组(P<0.05);Bad蛋白表达高于对照组(P<0.05)。结论:实验性肾阳虚大鼠生殖细胞凋亡明显增多,可能与Bcl-xl蛋白表达降低、Bad蛋白表达升高相关。生殖细胞凋亡可能是肾阳虚所致生殖功能障碍的一个重要原因。
Objective: To investigate the changes of germ cell apoptosis and the expressions of Bcl-xl and Bad in rats with experimental kidney-yang deficiency. Methods:12 male SD rats were randomly and equally allocated into a control and a kidney-yang deficiency group. The germ cell apoptosis was detected by TUNEL staining, and the expressions of Bcl-xl and Bad by immunohistochemistry. Results: Compared with the control rats, the apoptosis index of germ cells was significantly increased ( P 〈 0.05 ), the Bcl-xl expression obviously decreased ( P 〈 0.05) and the Bad expression markedly elevated ( P 〈 0.05) in the kidney-yang deficiency group. Conclusion: Lowered expression of Bcl-xl and lifted expression of Bad are associated with germ cell apoptosis in rats with experimental kidney-yang deficiency. Germ cell apoptosis may be a key reason for reproduction dysfunction induced by kidney-yang deficiency.
