摘要
从含有4"-酰化酶基因的螺旋霉素产生菌-基因工程菌的培养物中分离到一组4"酰化螺旋霉素,命名为生技霉素。其在体内对小鼠实验感染的治疗作用比乙酰螺旋霉素强。生技霉素主要组份为生技霉素A和B。通过溶媒提取,真空层析,制备薄层层析等方法得到了生技霉素A、B的纯品。生技霉素A的紫外最大吸收在230.8nm。具有与螺旋霉素、麦迪霉素、吉他霉素类似的红外吸收光谱,分子量为982(FABMS:M+H,983)。分子式C51H86N2O16。通过对FABMS,1H-NMR,13C-NMR,DEPT,COSY及HETCOR谱分析,鉴定生技霉素A的结构为4"-异戊酰螺旋霉素Ⅲ,并对4"-异戊酰螺旋霉素Ⅲ的84个连碳氢的1H-NMR信号作出了明确归属。生技霉素B与生技霉素A具有类似的紫外及红外吸收光谱。分子量为968(FABMS:M+H,969),比A小14个质量单位。主要根据1H-NMR及13C-NMR谱的分析及与生技霉素A的1H-NMR及13C-NMR谱的比较,鉴定生技霉素B的结构为4"-异戊酰螺旋霉素Ⅱ。
A group of 4' acyl spiramycins were isolated from the culture broth of spiramycin producing strain containing 4' acyl transferase gene, and were designated as shengjimycins. The shengjimycin complex showed a better in vivo activity than that of acetyl spiramycin. With the aid of vacuum liquid chromatography, centrifugal TLC and preparative TLC, pure shengjimycin A and B, the major components of shengjimycins, were obtained. Shengjimycin A has a maximum absorption at 230.8nm, and shows a similar IR spectrum to those of spiramycins, midecamycins and kitasamycins. Its molecular weight is 982 (FABMS: M+H, 983). Molecular formula is C 51 H 86 N 2O 16 . Based on FABMS, 1 H NMR, 13 C NMR, DEPT, COSY, HETCOR data the structure of shengjimycin A has been identified as 4' isovaleryl spiramycin Ⅲ. Based on careful analysis of 1 H NMR, COSY, and HETCOR spectrum, the signals of protons bonded to carbons were fully assigned. Sheng jimycin B has a similar UV and IR spectra to those of shengjimycin A. Its molecular weight is 968 (FABMS: M+H, 969). Based on the analysis of 1 H and 13 C NMR, the structure of shengjimycin B has been identified as 4' isovaleryl spiramycinⅡ.
