摘要
目的:建立以反相高效液相色谱法测定山楂叶提取物中槲皮素含量的方法。方法:色谱柱为Discovery C18(250mm×4.6mm,5μm),流动相为甲醇-0.4%磷酸,梯度洗脱,流速为0.6mL·min,柱温为(30±1)℃,检测波长为256nm。结果:槲皮素进样量在0.084-0.420μg范围内与峰面积积分值呈良好线性关系(r=0.9994,n=6)。结论:本方法操作简便、结果准确、重现性好,可用于山楂叶的质量控制。
OBJECTIVE: To determine the content of Quercetin in the leaves of Crataegus pinnatifida Bge.by RP- HPLC. METHODS: The chromatographic column was Discovery C18(250 mm × 4.6 mm,5 μm) with mobile phase consisted of methanol - 0.4 % phosphoric - acid in gradient elution at a flow rate of 0.6 mL · min ^-1.The column temperature was(30 ± 1) ℃and the detection wavelength was set at 256 nm. RESULTS: The linear range of Quercetin was 0.084-0. 420 μg( r = 0.999 4, n = 6) .CONCLUSION: The method is simple, accurate, reproducible, and applicable for the quality control of the leaf of Crataegus pinnati fida Bge..
