固相萃取/高效液相色谱法对中成药和保健品中7种降糖化学药物的测定 被引量:25

Simultaneous Determination of 7 Anti-diabetic Drugs in Traditional Chinese Medicines and Health Foods by Solid-phase Extraction and HPLC

摘要 建立了一种固相萃彬高效液相色谱测定保缝品中格列吡嗪、格列齐特、格列本脲、格列喹酮、盐酸二甲双胍、盐酸苯乙双胍和瑞格列奈7种降糖化学药物的方法。采用阳离子固相萃取柱,对提取液进行净化处理。得到的不同洗脱液分别进样,用液相色谱进行分析,采用ZORBAXSB—C18柱,以乙腈-0.02mol/L磷酸缓冲盐(pH2.8)作为流动相进行梯度洗脱,7个色谱峰分离良好,两次进样在40min内可以完成,外标法定量。在3种不同剂型中药制剂中的添加回收率在83%~101%之间。 An HPLC method for the determination of 7 anti-diabetic drugs glipizide, gliclazide, glibenclamide, gliquidone, repaglinide, metformin and phenformin in health foods was established. Maximum cation-exchange cartridge was used for sample treatment. The eluting solvent was optimized for the anti-diabetic drugs. Separations of analytes on a ZORBAX SB-C18 column(4.6 mm × 150 mm, 5 μm) were achieved by gradient elution at flow rate of 1 mL/min at ambient temperature with phosphate buffer(0.02 mol/L, pH 2. 8) and acetonitrile as mobile phases. The recoveries of 2 standard addition in three samples were 83% - 101% .
