摘要
目的:观察柠檬提取物对实验性小鼠急性柯萨奇B3m病毒性心肌炎的影响。方法:选取4~6周龄雄性Balb/C鼠腹腔注射柯萨奇病毒B3m株(CVB3m)建立实验性小鼠急性病毒性心肌炎模型。小鼠随机分为正常对照组、模型对照组及3个实验组(实验Ⅰ~Ⅲ组),每组10只。模型对照组和各实验组小鼠腹腔注射病毒1h后灌胃给药,实验Ⅰ~Ⅲ组分别灌服20、60、180倍稀释的柠檬提取液,模型对照组灌服等体积生理盐水,连用7d,正常对照组自然喂养,于第14天处死小鼠。观察各组小鼠心脏质量与体质量比(HM/BM)及心肌HE染色切片病理变化,并测定血清心肌酶含量。结果:(1)各实验组HM/BM较模型对照组明显降低(P<0.05),实验Ⅰ组、Ⅱ组心肌病理积分较模型对照组明显降低(P<0.05)。(2)实验Ⅰ组、Ⅱ组血清心肌酶活性较模型对照组明显降低(P<0.05或P<0.01)。结论:柠檬提取物对实验性小鼠急性柯萨奇B3m病毒性心肌炎具有一定的治疗作用。
Objective: To investigate the effect of lemon extraction on Coxsackievirus B3m (CVB3m) experimental acute myocarditis in mice. Methods: The Balb/C mice aged 4-6 w were chosen to establish the models of viral experimental acute myocarditis by inoculation intraperitoneally(i.p. ) with CVB3m. The mice were divided randomly into five groups( 10 for each group) which were normal control, viral control and three experimental groups (Group Ⅰ-Ⅲ ). Mice in experimental groups were gastric incubated the lemon extraction lh after infection of CVB3m. The concentrations administrated by the mice in each experimental groups were 20-fold-diluted of lemon extraction for Group Ⅰ ,60-fold-diluted for Group Ⅱ , and 180- fold-diluted for Group Ⅲ. And mice in viral control were gastric incubated saline at the same time. The treatment of lemon extraction lasted 7 d. The mice in normal control were not taken any special treatment. Then the mice in all groups were sacrificed on the 14th day after CVB3m infection. The value of heart to body mass ratio (HM/BM) and pathological change of myocardium were observed. Also the concentrations of plasma cardiac enzymes were measured. Results: The values of HM/BM in 3 experimental groups were significantly decreased compared to that in the viral control (P 〈 0.05). The values of myocardial pathology and the concentrations of serum cardiac enzymes in experimental Ⅰ and experimentalⅡ groups were lower than that in viral control (P 〈 0.05 or P 〈 0.01 ). Conclusion: The lemon extraction has the therapeutic effect on CVB3m experimental acute mvocarditis in mice.
