摘要 学术背景:近年来研究发现,羊水细胞中含有多种干细胞的标记物,且在体外通过特殊的诱导微环境能够分化成多种不同类型的细胞,这使羊水来源干细胞成为干细胞研究的一个新的热点。目的:对各种羊水来源干细胞的特征、培养方法和分化能力加以综述,并介绍其应用前景。检索策略:应用计算机检索Pubmed数据库1991-01/2007-08期间相关文献,检索词为"amniotic fluid cells,stem cells",限定语言种类为English。对资料进行初审,选取有关羊水来源干细胞的基础与临床研究,排除明显不相关的研究和综述文献。文献评价:共收集到106篇相关的文献,31篇符合要求。其中基础性研究有18篇,利用动物模型研究的文献有13篇。2篇关于羊水细胞的背景介绍,9篇关于羊水细胞的组成,8篇关于羊水细胞的分离培养与鉴定,1篇关于羊水细胞的分化能力,8篇关于羊水来源干细胞的应用前景,3篇关于相关问题与展望。资料综合:羊水中包含许多已分化的和未分化状态的细胞,已被证实间充质干细胞等多能干细胞存在于羊水中,羊水细胞还表达神经干细胞的表面标记和端粒酶及Oct-4等多(潜)能干细胞的表面标记。可以采用单抗贴壁铺展法、流式细胞分选法、免疫磁珠分选法等分离培养羊水干细胞。由于首次从羊水分离获得的干细胞数目往往很少,因此需要在体外对分离出来的干细胞进行非分化性培养扩增。目前已经证实羊水来源的干细胞具有多向分化能力,因此羊水作为干细胞的一个新的来源,将为干细胞的研究揭开一个新的研究领域,为组织工程提供新的种子细胞来源。结论:羊水来源干细胞的研究尚处于初始阶段,它的许多生物学特性还不清楚。 BACKGROUND: There are many markers of stem cells in amniotic fluid, and the stem cells from amniotic fluid can differentiate into various cells under specific induced environment in vitro. Accordingly, the study of the stem cells from amniotic fluid has become one new focus in this field. OBJECTIVE: To review the characterization, culture methods and differentiated abilities of the stem cells from amniotic fluid, and introduce the application of these cells. RETRIEVAL STRATEGY: A computer-based online search of Pubmed was undertaken for English full-text articles published from January 1991 to August 2007 with keywords "amniotic fluid cells, stem cells". After the first trial, only basic research and clinical studies about amniotic fluid-derived stem cells were selected. Obvious unrelated researches and review articles were excluded. LITERATURE EVALUATION: 106 related articles were collected, after removing the unrelated studies, 31 articles were included. Of the 31 articles, 18 were basic researches, and 13 were studies by animal model; 2 were about introduction of amniotic fluid cells, 9 about composition of amniotic fluid cells, 8 about isolation, culture and identification of amniotic fluid cells, 1 about differentiation ability of amniotic fluid cells, 8 about application prospect of amniotic fluid-derived stem cells, and 3 about the problems and future tendency. DATA SYNTHESIS: Many undifferentiated and differentiated cells have been found in human amniotic fluid. It has been documented that amniotic fluid contains multipotent stem cells, such as mesenchymal stem cells. In addition, it is described that amniotic fluid cells express markers for neuronal stem cells and many markers for pluripotent stem cells, such as Telomerase, Oct-4, et al. The stem cells in amniotic fluid can be obtained by monoclonal antibody, flow cytometry and immunoselection with magnetic microspheres. The stem cells must be cultured and amplified under undifferentiated because the firstly acquired stem cells from amniotic fluid are few. It has been validated that amniotic fluid-derived stem cells has multi-differentiation capability. Therefore, amniotic fluid as novel resource for stem cells will open a new field in stem cell research and provide a novel seed cell in tissue engineering. CONCLUSION: The investigation of stem cells from amniotic fluid is just onset, and the characteristics of the stem cells are not clear yet.
