摘要
目的:建立链脲佐菌素(STZ)诱导的糖尿病大鼠模型并用霉酚酸酯(MMF)干预,动态观察细胞因子(RANTES)、单核/巨噬细胞表面特异性标志抗原(ED-1)、Ⅳ型胶原(ColⅣ)在肾组织中的表达,探讨MMF能否通过抑制肾组织炎症反应达到保护肾脏的作用。方法:36只雄性SD大鼠随机分为正常对照组、糖尿病模型组和MMF治疗组,于实验第4周、14周末每组分别处死6只大鼠,处死前留取24h尿进行24h尿蛋白定量,处死后免疫组织化学染色观察RANTES、ED-1、ColⅣ在肾组织中的表达。结果:(1)正常肾组织RANTES有少量表达,模型组的表达均显著增高,MMF组各时间点与相应模型组比较均显著降低;(2)正常肾组织ED-1有少量表达,模型组的表达均显著增高,MMF组各时间点与相应模型组比均显著降低。结论:MMF可能是通过下调RANTES在肾组织中的表达、减少单核/巨噬细胞在肾组织中的浸润,在早期抑制肾组织炎症反应,进而对肾脏具有一定的保护作用。
Objective:To observe the expression of RANTES, ED-land Col IV in the renal tissue of diabetic rats intervened by MMF, and explore whether MMF can inhibit inflammatory reaction to protect kidney. Methods:Thirty- six male Sprague - Dawley rats were randomly divided into three groups after uninephrectomy: normal control group, diabetic model group and MMF -treated group. Six rats of each group were sacrificed at the 4th week and 14th week after STZ injection. 24 hours urinary protein was measured before the rats were sacrificed. The expression of RANTES,ED- land Col IV protein in renal tissue were detected by immunohistochemistry. Results: ( 1 )The expression of RANTES was little in normal renal tissue. In diabetic rnodel group, it was obviously higher than that in the normal control group at each time point. It was decreased after treatment with MMF at each time point. (2)The expression of ED - 1 was little in normal renal tissue. In diabetic model group, it was obviously higher than that in the normal control group at each time point. It was decreased after treatment with MMF at each time point. Conclusion: MMF may play a certain role in renoprotection in the diabetic model, and it may be through reducing the expression of RANTES protein and histoleucocyte/macrophagus infiltrating in renal tissue, and through inhibiting inflammatory reaction in the early stage.
