应用siRNA敲低MMP-9基因表达后对人U251胶质瘤细胞增殖和侵袭的作用 被引量:9

摘要 目的研究siRNA靶向抑制MMP-9基因对U251细胞的增殖和侵袭的抑制作用。方法采用oligofectamine转染试剂将siRNA导入到胶质瘤细胞U251,分别采用半定量PCR及Western blotting从mRNA和蛋白水平检测干扰效果;采用细胞生长曲线、MTT和流式细胞法检测siRNA对细胞增殖的影响,应用2D、3D生长实验(Two,three-dimensional growth experiment)和Transwell实验检测转染MMP-9 siRNA后对细胞侵袭的影响。结果半定量PCR以及Western blotting显示MMP-9基因的mRNA转录水平以及蛋白水平的表达均得到抑制;细胞生长曲线、MTT检测及流式细胞仪检测结果显示MMP-9基因表达被抑制后,U251细胞增殖率明显受到抑制,2D、3D生长实验和Transwell侵袭实验证实转染后U251细胞体外侵袭能力明显降低(P<0·01)。结论应用siRNA敲低胶质瘤细胞MMP9基因表达后,胶质瘤细胞的侵袭和增殖能力明显下降。
