摘要
将聚苯乙烯作为载体,以氯乙酰化代替氯甲基化,制备了聚苯乙烯酰乙基苯丙氨酸-Ni2+树脂(PS-Phe-Ni2+)。该树脂合成方法简单,过程中不使用致癌物氯甲醚等;羰酰乙基可与苯丙氨酸上的氧与Ni2+形成2个五元环,使该树脂对Ni2+的螯合稳定且螯合量较高,有利于提高蛋白吸附量。以醛糖还原酶融合蛋白为对象,初步考察了该树脂在纯化融合蛋白中的应用。在合适的条件下纯化倍数可达30倍。
A polystyrene acetyl phenylalanine-Ni^2+ resin (PS-Phe-Ni^2+) is prepared by chloroacetylated polystyrene instead of chloromethyl polystyrene resin as the matrix, This process is simple and avoids carcinogenic compounds ( such as chioromethyl ether, etc. ) .The carbonyl of PS-Acyl-Cl and oxygen atom of L-Phe can form two five-membered rings with Ni^2+ to improve the adsorption capacity and stabilize the chelation of the resin for Ni^2 + , which is favorable for the adsorption capacity of protein. PS-Phe-Ni^2+ is used to purify aldose reductase fusion protein, and under optimal conditions the protain can be purified up to 30 folds from the original.
