摘要
目的建立血清总胆汁酸(TBA)测定的酶循环法。方法在全自动生化分析仪上用3α羟类固醇脱氢酶和辅酶体系循环底物的原理,建立循环酶测定法。结果本法批内、批间精密度结果(μmol/L)及变异系数分别是7.31±0.23、2.59%,7.38±0.282、3.82%;线性达100μmol/L;与商品试剂盒比较r=0.998;平均回收率97.7%;在2-8℃避光可稳定一个月。对90例健康人和85例不同肝脏疾病血清TBA进行测定,健康人参考范围在3.0-10.0μmol/L,可敏感的区分出不同肝脏疾病时血清胆汁酸的含量变化,符合临床需求。结论本法在全自动生化分析仪上采用循环酶速率法测定血清TBA,具有速度快,简便,灵敏度高,线性、精密度好的优点,是一种理想的TBA测定方法。
Objective Establishing a circulating enzymatic method, which is used to detect serum total bile acid(TBA). Methotis Utilize the principle of 3a-HSD and coenzyme systerm cycling substrate to establish the circulating enzymatic method by using the automated biochemistry analysis. Results The precision results and the coefficient of variation of the within - run and between- run are 7.31± 0.23,2.59 %, 7.38 ± 0.21,2.62 % ; this method had a linearity up to 180wnol/1; compared with commercial Kit, its r = 0. 998 ; the average recovery is 97.7 % ; The 3a-HSD can be steadly stored in the dark condition, under 2 - 8 ℃. The serum TBA of healthy group(n = 90)and defferent types of hepatic diseases group were detected,the values of the healthy group are between 5.0 wnol/1 and 10.0 wnol/1,and this method can sensitively identify the clinical value of serum TBA in different types of hepatic diseases, which meets for clinical demand. Conclusion The circulating enzymatic method of detecting serum TBA by using the automated biochemistry analys is is of some advantages, like high speed, simlpe instrument, high sensitivity, fine linear and precision. Therefore, it is an ideal method of detecting the serum TBA.
