摘要
目的:研究钒化钠对小鼠成纤维细胞NIH3T3细胞株细胞周期蛋白(Cyclin)D1表达的影响,探讨钒化钠在细胞生长中的作用机制。方法:以体外培养小鼠成纤维细胞NIH3T3细胞株为研究对象,给予0、0.1、1.0、5.0、10.0、100.0、200.0μmol/L钒化钠,培养4小时后,用Western blot技术检测小鼠成纤维细胞NIH3T3细胞株Cyclin D1表达情况。结果:5.0、10.0μmol/L钒化钠组培养4小时后,Cyclin D1蛋白表达均增高;100.0、200.0μmol/L钒化钠组培养4小时后,Cyclin D1蛋白表达均降低。结论:钒化钠可通过上调和下调Cyclin D1的蛋白表达的双重作用,来影响细胞的生长。
Objective : To study the effect of sodium orthovanadate on the expression of cell cycle protein Cyclin D1 in mouse NIH3T3 cell line. Methods: Mouse NIH3T3 cells grown in cell culture were treated with sodium orthovanadate at doses of 0, 0.1, 1.0, 5.0, 10.0, 100.0,200.0μmol/L respectively for 4 hours and the cell extracts were subjected to Western blotting technique to examine the expression of cyclin D1. Results: After application for 4 hours, as compared with the control, sodium orthovanadate at the dose of 5.0 or 10.0μmol/L induces the expression of Cyclin D1 significantly whereas at the dose of 100.0 or 200.0μmol/L, it causes the reduction of cyclin D1 expression. Conclusion: Sodium orthovanadate may affect the growth of NIH3T3 cells through the bi - directional control of cyclin D1 expression including both down - regulation and up - regulation.
