白细胞介素-12和干扰素-γ表达与变应性鼻炎的相关性 被引量:6

Expression of IFN-γ and IL-12 in patients with allergic rhinitis

摘要 目的探讨变应性鼻炎患者白细胞介素12(interleukin-12,IL-12)和干扰素-γ(interferon-gamma,IFN-γ)的表达及两者与变应性鼻炎的相关性。方法通过原位杂交染色法和酶联免疫吸附剂测定(enzyme-linked immunosorbnent assay,ELISA),检测30例变应性鼻炎患者和22例对照鼻黏膜组织中IL-12和IFN-γ的表达。结果在变应性鼻炎组和对照组,外周血单个核细胞(peripheral blood mononuclear cell,PBMC)IL-12mRNA杂交阳性率分别为0.13±0.03和0.77±0.15(t=11.1,P<0.05);血清IL-12的含量为(13.7±2.4)pg/L和(52.9±8.4)pg/L(t=6.2,P<0.05);血清IFN-γ的含量为(11.2±2.1)pg/L和(51.8±6.4)pg/L(t=7.9,P<0.05)。结论变应性鼻炎的病理过程与IL-12和IFN-γ的表达下调有关。Th细胞分化偏移,Th2功能亢进是导致变应性鼻炎的重要原因。 OBJECTIVE To explore the relationship between allergic rhinitis and IFN-γ, IL-12. METHODS The expression of IL-12 was detected by in situ hybridization and enzyme-linked immunosorbent assay in 37 patients with allergic rhinitis and 12 control samples. IFN-γ was detected by ELISA. RESULTS The positive ratios of IL-12 in PBMC were 0.13±0.03 and 0.γγ±0.15 in allergic rhinitis group and control group respectively. The concentrations of IL-12 were (13.7±2.4) pg/L and (52.9±8.4) pg/L in allergic rhinitis group and control group respectively, The concentrations of IFN-γ were (11.2 ± 2.1 ) pg/L and (51.8 ± 6.4) pg/L in allergic rhinitis group and control group respectively. CONCLUSION The down-regulation of IL-12 and IFN-γ was related to allergic rhinitis, and hyper-function of Th2 cells may play an important role in the pathogenesis of allergic rhinitis.
