摘要 建立一种快速、稳定、灵敏并可反映细菌活性的细菌计数方法。以乳酸菌中的保加利亚乳杆菌和嗜热链球菌为研究对象,探讨MTT法用于细菌计数的可行性及测量细菌数量的范围、反应时间、MTT的用量、是否加入溶解剂等实验条件。结果:保加利亚乳杆菌在(1.0×105~2.18×107)cfu/mL内测出的OD570值与细菌浓度呈良好的正相关,反应时间1.5h,MTT添加量20μL,测量前用DMSO溶解;嗜热链球菌在(2.0×105~5.12×107)cfu/mL范围内测出的OD570值与细菌浓度呈良好的正相关,反应时间2.0h,MTT添加量20μL,可不添加溶解剂直接测量。MTT比色分析法可用于检测乳酸菌活菌数量。 To establish a rapid, steady and sensitive bacteria-counting method that could reflect the bacteria's activity. Lactabacillus delbrueckii ssp. bulgaricus and St reptococcus thermophilus were employed to discuss the feasibility of application of MTT method to bacteria-counting and determine the conditions of the assay, including linear range, reaction time, MTT dosage and whether solvent is added etc.. Results: The optical density at 570 nm is positively related to the concentration of L,delbrueckii ssp. bulgaricus when the number of the bacteria is in the range of (1.0× 10^5-2.18× 10^7) cfu/mg, and reacting time is 1.5 h, MTT dosage is 20 μL, and DMSO is used as solvent before assay; For St.thermophilus, the optical density at 570 nm is positively related to the concentration of the bacteria when the number of the bacteria is in the range of (2.0× 10^5-5.12× 10^7) cfu/mL, and reacting time is 2.0 h, MTT dosage is 20 μL and solvent is not added.. Conclusion: MTT colorimetry could be used to measure the viability of lactic acid bacteria.
