摘要
目的探讨NF-κB、PCNA、VEGF在胰腺癌组织中的表达关系及其意义。方法用免疫组织化学法检测正常胰腺组织、胰腺癌癌旁组织及癌组织中NF-κB、PCNA、VEGF的表达,并分析其与胰腺癌临床病理特征的相关性。结果胰腺癌组织中NF-κB、VEGF、PCNA的阳性表达率分别为64.6%、63.1%、81.5%,显著高于正常胰腺组织和癌旁不典型增生组织的表达率。NF-κB的表达与肿瘤直径、肿瘤分化程度、TNM分期、淋巴结转移有关(P<0.05),而与性别和神经浸润无关(P>0.05);PCNA的表达与肿瘤直径、淋巴结转移有关(P<0.05),而与性别、组织分化程度、神经浸润、TNM分期无关(P>0.05);VEGF的表达与肿瘤分化程度、TNM分期、淋巴结转移有关(P<0.05),而与性别、肿瘤直径、神经浸润无关(P>0.05)。结论NF-κB、PCNA、VEGF在胰腺组织中的阳性表达率随组织异形性增加而升高,NF-κB、PCNA、VEGF表达之间呈正相关,NF-κB可能通过正向调节PCNA、VEGF的表达促进胰腺癌的发生发展。
Objective To explore expression and the significance of of nuclear factor- kappaBp65 (NF- κB p65), proliferating cell nuclear antigen(PCNA) and vascular endothelial growth factor(VEGF) in pancreatic cancer. Methods The expression of NF- κB,VEGF and PCNA in normal pancreatic tissues,atypical hyperplastic tissues and pancreatic cancer tissues were detected by immunohistochemistry (SP),and clinic pathological correlation with pancreatic cancer was analyzed. Results The positive exression rates of NF - κB, VEGF and PCNA in pancreatic cancer were 64.6% ,63.1% ,81.5% respectively.The expression rate in pancreatic cancer were higher than those in normal pancreatic tissues and atypical hyperplastic tissues. It was found that the expression of NF- κB was in significant relation to diameter of tumor, tumor cell differentiation, TNM stages and lymph node metastasis( P 〈 0.05), but no relation to gender and perineuml invasion; that the expression of PCNA was in significant relation to diameter of tumor and lymph node metastasis( P 〈 0.05),but no relation to gender, tumor cell differentiation, perineuml invasion and TNM stages; that the expression of VEGF was in significant relation to tumor cell differentiation, TNM stages and lymph node metastasis ( P 〈 0.05), but no relation to gender, diameter of tumor and perineuml invasion. Conclusion The positive expression rares of NF - κB, PCNA, VEGF in pancreatic tissues increased with the escalating severity of tissue atypia. The expression of NF- κB, PCNA and VEGF were positively correlated with each other, which indicated that NF - κB possibly upregulated the expression of PCNA and VEGF during carcinogenesis and accelerated the progression of pancreatic cancer in cooperation with them.
