酸枣仁皂苷A对大鼠局灶性脑缺血再灌注后谷氨酸免疫组化阳性细胞及神经行为学的影响 被引量:6

摘要 目的观察酸枣仁皂苷A对大鼠脑缺血再灌注后谷氨酸免疫组化阳性细胞及神经行为学的影响。方法96只SD大鼠随机分为假手术组、缺血再灌注模型组、尼莫地平组和酸枣仁皂苷A组,除假手术组外其余各组分设12 h、24 h、48 h、4 d、7 d五个时间组。参考Longa线栓法制备大脑中动脉闭塞再灌注损伤模型,造模后分组给药并按时点对大鼠进行神经行为学评分后断头法处死动物,取脑组织切片作谷氨酸免疫组化染色。结果造模后12 h缺血脑组织中心区可见大量谷氨酸阳性细胞,48 h达高峰,7 d后明显减少;尼莫地平及酸枣仁皂苷A组谷氨酸阳性细胞数量较模型组明显减少,神经功能缺失症状亦较模型组有所减轻。结论酸枣仁皂苷A能抑制脑组织谷氨酸免疫组化阳性细胞的表达,改善缺血再灌注损伤大鼠神经行为缺失症状,在脑缺血急性期具有脑保护作用。 Objective To study the effects of Jujuboside A on neuroethology and Glutamate immunohistoehemieally positive cells in cerebral ischemia-reperfusion injury rats. Methods Ninety-six SD rats were randomly divided into four groups: sham group, cerebral ischemiareperfusion model group, Nimodipine group and Jujuboside A (JuA) group. Except the sham group, the other groups were divided into 5 time-groups, as 12 h, 24 h, 48 h, 4 d, and 7 d. The models of middle cerebral artery occlusion (MCA0) were established by using Zea Longa's method. Each group was injected with related medicine after modeling and graded the neurological behavior on time-points. Then, the brains were removed on each time-point after modeling and sliced up for Glutamate (Glu) immunohistochemical cell detection. Results Twelve hours after modeling, a lot of Glutamate immunohistochemical cells were found in the isehemic core regions, which reached peak at 48h and reduced obviously after 7 d. Compared with model group, the quantities of Nimodipine group and JuA group obviously decreased and the symptoms of rats' neurological deficit were also alleviated. Conclusion JuA may decrease the expression of Glutamate immunohistochemically positive cells and improve the symptoms of rats' neurological deficit, which has some protective effects on the brain in acute cerebral ischemia.
