SIRS和MODS防治新对策的实验研究——血必净的药效学观察 被引量:165

摘要 目的:探寻既有拮抗内毒素作用,又有拮抗炎性介质释放作用的复方中药。方法:从36组中药复方中筛选出由赤芍等5味中药组成的血必净静脉用针剂,观察体外法血必净抗内毒素药效、体内法血必净抗炎性介质药效及血必净对实验性内毒素性全身炎性反应综合征(SIRS)和多脏器功能失常综合征(MODS)动物模型存活的保护作用。结果:血必净有强效的拮抗内毒素作用(血必净含生药浓度0.150g/L,37℃温浴1小时即有拮抗内毒素作用;含生药0.075g/L,37℃温浴4小时即有拮抗内毒素作用),血必净也有强效的抑制TNFα释放的作用;血必净能明显提高实验性SIRS和MODS动物模型的存活率(SIRS和MODS模型非治疗组的动物存活率16.7%,SIRS和MODS加血必净组存活率100.0%)。结论:血必净疗法为提高临床防治SIRS和MODS的疗效提供了可借鉴的新对策。 Objective:To seek an effective traditional Chinese herb prescription,which possesses the effects of both antiendotoxin activity and antiinflammatory mediators release,in the treatment of systemic inflammatory response syndrome (SIRS) and multiple organ dysfunction syndrome (MODS).Methods:From 36 groups traditional Chinese herb prescriptions,we selected one effective compound prescription consisting of 5 kinds of herbs including paeonia ruba.It was called “xue bi jing”,and it was prepared for intravenous use.Both in vitro and in vivo experiments were performed to observe the effects of “xue bi jing” on inflammatory mediators release and survival rate in animals with SIRS and MODS induced by endotoxin challenge.Results:“Xue bi jing” had strong effects on blockade of endotoxin,showing antiendotoxin activity in a dose of 0 150 g/L incubated at 37 ℃ for an hour or 0 075 g/L at 37 ℃ for 4 hours.Also,it could significantly inhibit the release of tumor necrosis factorα.Moreover,treatment with “xue bi jing” showed protective effect in animals with SIRS and MODS (survival rate,nontreated group vs.treated group:16 7% vs.100 0%).Conclusions:These data indicate that “xue bi jing” is a new drug in the prevention and treatment of SIRS and MODS.
