正交试验法优选超声提取河豚毒素的研究 被引量:4

摘要 目的:研究从河豚鱼内脏中提取河豚毒素(TTX)的最佳提取条件。用超声提取法与传统的水浴法和回流法进行比较。方法:采用超声提取法对河豚毒素的提取方法进行研究,设计L9(34)正交表,采用紫外分光光度法检测,以提出的TTX含量为评价指标,优选TTX的提取条件。结果:超声时间、乙酸用量、提取次数对TTX的提取量均有影响,其中提取时间有显著影响,最佳提取工艺为用河豚内脏的3倍量的0.1%的乙酸,超声提取3次,每次40min。超声提取法优于水浴法和回流法。 Objective: To choose the optimal method of extracting tetrodotoxin(TTX) from puffer -fish intemal organs. We have compared the result of ultrasonic extraction method with of the method of immersion or backstreaming. Methods: ultrasonic extraction was used to the extraction process of tetrodotoxin ,design L9 (3^4) orthogonal chart to optimize the extraction of TTX, and the detection of tetrodotoxin was determinedby ultraviolet spectrophotometry with content of tetrodotoxin as markers. Results : ultrasonic time, acetic consumption, extraction times have effect on the content of extracting tetrodotoxin, And the influence of ultrasonic time was significant. The optimal extraction process was 0.1% acetic acid ,3times the amount of material, ultrasonic extraction for 3 times ,40 minutes each time. The ultrasonic extraction method was markedly superior to the method of immersion and backstreaming.
