迷迭香酸对慢性肾功能不全大鼠系膜基质及TGF-β_1的影响 被引量:4

Effects of Rosmarinic Acid on the Glomerular Expresion of Mesangium Matrix and TGF-β_1 in Rat Model of Chronic Renal Failure

摘要 目的观察迷迭香酸对慢性肾功能不全模型大鼠系膜基质及转化生长因子-β1(TGF-β1)的影响。方法制作大鼠慢性肾功能不全模型(5/6肾切除),以迷迭香酸作为干预因素,应用免疫组织化学半定量法测定迷迭香酸对大鼠系膜基质成分纤维连接蛋白(Fn)、胶原Ⅳ(Col-Ⅳ)及TGF-β1的影响,电镜观察肾小球病理学变化,测定血、尿肌酐值。结果(1)与对照组比较,迷迭香酸组肌酐清除率(Ccr)水平升高(P<0.05);(2)Fn、Col-Ⅳ在肾脏细胞外基质的沉积减少(P<0.05);(3)TGF-β1表达减少(P<0.05)。结论迷迭香酸可以抑制TGF-β1的表达,减少Fn及Col-Ⅳ在肾小球细胞外基质的沉积,延缓肾小球硬化,对慢性肾功能不全疾病可能具有治疗价值。 Objective To explore the effects of rosmarinic acid on the glomerular mesangium matrix and the expresion of transforming growth factor beta1 (TGF-β1) of the 5/6 nephrectomy Sprague-Dawley(SD) rats. Methods The animal model of chronic renal failure was first established. The effects of rosmarinic acid on the glomerular mesangium matrix and the TGF-β1 expresion of the 5/6 nephrectomy SD rats were delected by immunohistochemistry. Electron microscope was used to examine the pathology of the renal constitution of the chronic renal failure rats. Results (1) Compared to the matched control group, the creatinine clearance rates of treated groups were ameliorated(P〈0.05). (2) The expresion of fibronectin and collagen-IV in the glomerular mesangium matrix was significantly decreased(P(0.05). (3) Rosmarinic acid decreased the expression of TGF-β1 in the kidney(P(0.05). Conclusion Rosmarinic acid suppressed the expression of TGF-β1 in the kidney and decreased the number of fibronectin and collagen-IV in the glomerular mesangium matrix. Rosmarinic acid might be a promising agent to treat chronic renal failure diseases.
