啤酒花中蛇麻酮诱导细胞凋亡的实验研究

摘要 目的观察啤酒花中成分蛇麻酮对肿瘤细胞SGC-7901和HepG-2诱导细胞凋亡的作用,并揭示其作用机制。方法采用MTT法观察蛇麻酮体外抗肿瘤作用,采用流式细胞仪观察蛇麻酮对肿瘤细胞周期及细胞凋亡的影响。结果蛇麻酮对肿瘤细胞SGC-7901及HepG-2均有较好的疗效。蛇麻酮作用于SGC-7901和HepG-2细胞50 h后,肿瘤细胞均有明显凋亡峰出现。结论蛇麻酮可以将肿瘤细胞SGC-7901和HepG-2的细胞周期阻滞在G0/G1和S细胞期,升高细胞凋亡指数。 Objective It is to observe the action of lupulone in humulus lupulus inducing apoptosis on tumor cell SGC- 7901 and HepG- 2 and to reveal the mechanism. Methods The anti-tumor effect of lupulone in vitro was observed with MTT method. The influence of lupulone on cell cycle and apoptosis of tumor cell was observed with flow cytometer. Results Lupulone had both better curative effects on tumor cell SGC - 7901 and HepG- 2. After lupulone affecting on SGC - 7901 and HepG - 2 for 50 h, tumor cells all had appeared obvious apoptosis peak. Conclusion Lupulone can block the cell cycle of tumor cell SGC - 7901 and HepG - 2 at G0/G1 and S phase and can heighten apoptosis index.
