摘要 研究了食品中胭脂虫红色素的高效液相色谱测定方法。含胭脂虫红色素的样品用液-液萃取后通过高效液相色谱进行分析,并通过比较测定的光谱与胭脂虫红的紫外-可见谱图库来进一步确证。采用Welch Materials XB-C18(4.6mm×250mm,5μm)柱,流动相为0.02mol/L乙酸铵溶液(A)和甲醇(B),梯度洗脱,流速为1.0mL/min,检测波长为280nm,外标法测定胭脂虫红的含量。结果表明,胭脂虫红在1.0~50.0mg/L范围内线性关系良好,回收率为88%~99%,检出限为0.041mg/L,色谱峰分离效果好,具有良好的稳定性和重复性。 A sensitive HPLC method for the analysis of carminic acid in food was described. Food samples were extracted by liquid - liquid extraction and chromatographied on a Welch Materials XB - C28 (4- 6 mm ×250 mm, 5 μm) column and detected at 280 nm, with a mobile phase of 0. 02 mol/L ammonium acetate solution -methanol( gradient elution) at a flow rate of 1.0 mL/min. The linearity was obtained in the range of 1.0 - 50. 0 mg/L ( r = 0. 999 9 ). The determination limit was 0. 041 mg/L. The average recoveries of samples was in the range of 88% - 99% . The analytical method for carminic acid in food was proved to be simple, sensitive and reliable.
