摘要
为研究干酪乳杆菌Lb.casei.Zhang细胞壁肽聚糖(peptidoglycan;PG)的抗肿瘤作用,用流式细胞仪检测肽聚糖对人胃癌BGC-823细胞生长周期的影响。以肝癌H22细胞移植瘤昆明小鼠为模型,分别采用原位杂交和免疫组化方法检测小鼠移植瘤增殖细胞核抗原(PCNA)mRNA的表达以及凋亡因子Bcl-2和Bax基因的蛋白表达。结果显示,肽聚糖能够阻滞人胃癌BGC-823细胞生长于G0/G1期;实验组PCNA mRNA的表达和Bcl-2基因的蛋白表达平均光密度值均明显低于对照组,而Bax基因蛋白表达情况则相反,与对照组比较,均差异显著(α=0.05),且抑瘤率达37.21%。提示干酪乳杆菌Lb.casei.Zhang细胞壁肽聚糖具有抗肿瘤作用。
To study effect of peptidoglycan (PG) of Lb. casei. Zhang cell wall against tumor , the cell cycles were analyzed in human gastric cancer BGC-823 cells using flow cytometry. The positive expression of the proliferating cell nuclear antigen (PCNA) mRNA. Bcl-2 and bax oncoprotein were investigated in mouse transplantation tumors by in situ hybridization and immunohistochemistry technique, depending on the animal model of Kunming mouse transplantation tumors of hepatoma H22. The results showed that: peptidoglycan could cause the G0/G1 phase block of BGC-823 cells. The mean optical density of PCNA mRNA and Bcl-2 oncoprotein in peptidoglycan group were significantly lower than those in the control group. The mean optical density of Bax was contrary to that of Bcl-2 oncoprotein. Compared with control group, the differences were all significant. The tumor inhibiting ratio of peptidoglycan was 37.21%. The good effect of peptidoglycan of Lb. casei. Zhang cell wall in vivo and in vitro indicated that it had anti-tumor activity.
