摘要
目的通过高血压脑出血肝阳化风证、高血压病肝阳上亢证及健康对照组外周血单个核细胞的双向凝胶电泳图谱的建立,鉴定并分析差异表达的蛋白质,以探讨肝阳上亢证与肝阳化风证证本质内涵。方法用淋巴细胞分离液分离出高血压脑出血肝阳化风证组、高血压病肝阳上亢证组及健康对照组外周血单个核细胞,采用二维凝胶电泳分离三组总蛋白质,获得差异表达蛋白质,制备蛋白质组表达图谱,PDQuest软件分析,并通过基质辅助激光解吸电离飞行时间质谱(MALDI-TOF)和数据库对差异表达的蛋白质分析鉴定。结果建立了高血压病肝阳上亢证,高血压脑出血肝阳化风证及健康对照外周血单个核细胞蛋白质表达图谱,经比较发现,肝阳上亢证与肝阳化风证有6个相同表达的蛋白质,18个差异表达的蛋白质。鉴定出的蛋白质涉及细胞骨架蛋白、代谢酶类、细胞信号转导蛋白、抗氧化蛋白、血液蛋白等。结论高血压病肝阳上亢证与高血压脑出血肝阳化风证具有相同和差异表达的蛋白质,提示高血压病肝阳上亢证和高血压脑出血肝阳化风证有共同的物质基础和不同的本质内涵,为"肝阳化风证"证本质的深入研究奠定了基础。
Objective To explore the essence of Hyperactivity of Liver Yang Syndrome （HOLYS） and Liver Yang Forming Wind Syndrome （LYFWS） by establishing the two dimensional polyacrylamide gel electrophoresis （2- DE） expression maps of total protein in peripheral blood mononuclear cells （ PBMCs） of the primary hypertension patients with HOLYS, the hypertension hemorrhage patients with LYFWS. and the norma persons, and analyzing the different expression proteins. Methods PBMCS of the hypertension intracerebral hemorrhage patients with LYFWS. the primary hypertension patients with HOLYS. and the normal persons were isolated by using lymphozytes separation medium. 2 DE technology was applied to separate the total proteins of the three groups to gain different expression protein, establish 2 - DE image, and analyze them by PDQuest 7.3.1 software. Different expression proteins were identified and analyzed by MALDI TOF - MS and data- base. Results The expression maps of proteins in PBMCS of primary hypertension patients with HOLYS. hypertension intracerebral hemorrhage patients with LYFWS. and normal persons were established. Eighteen different protein spots and 6 identical protein spots were found in HOLYS group and LYHFS group. The identified proteins include cytoskeletal proteins, metabolic enzymes, signal transmission proteins, antioxydation proteins, blood proteins and so on Conclusions The HO- LYS and the LYHFS have identical proteins and different proteins, which mean that HOLYS and LYHFS have same matter foundation, and have different essence. This found may lay a foundation for the further investigation of essence of the LYFWS.
