摘要
目的:研究玉泉丸(葛根、天花粉、地黄等)对盐酸二甲双胍在大鼠体内药动学过程的影响。方法:大鼠灌胃给予玉泉丸7d后,给予盐酸二甲双胍,给药后不同时间采集血样。用高效液相色谱法测定盐酸二甲双胍血药浓度,绘制药-时曲线,用DAS1.0统计软件进行处理,获得药动学参数。结果:二甲双胍组、联合用药组的主要药动学参数分别为:Cmax=8.57μg/mL、t1/2=232.8min、Cl/F=0.092(L/min)/kg、AUC=3291.5mg/L*min;Cmax=11.60μg/mL、t1/2=523.9min、Cl/F=0.045(L/min)/kg、AUC=5019.7mg/L*min。结论:盐酸二甲双胍在大鼠体内药动学过程符合一室模型,玉泉丸对盐酸二甲双胍在大鼠体内的药动学有显著影响。
AIM: To study the effects of co-administration Yuquan Pill (Radix Puerariae lobatae, Radix Tricho-santhis, Radix Rehmanniae, etc. ) on the pharmacokinetics process of metformin hydrochloride. METHODS: After administering Yuquan Pill for seven days, the metformin hydrochloride was orally administered to the rats, then the blood samples were collected at different time. The concentrations of metformin hydrochloride in plasma were determined by HPLC method. According to the curve of concentration-time, the software of DAS1.0 was used to process the pharmacokinetic parameters. RESULTS: The main pharmacokinetic parameters of the controlling group and the testing group were C max = 8.57 μg/mL,t1/2 = 232.8 min,C1/F = 0. 092 (L/min)/kg,AUC = 3 291. 5 mg/L * min; Cmax, = 11.60 μg/mL,tl/2 =523.9 min ,C1/F =0. 045 ( L/min)/kg,AUC =5 019.7 mg/L * min, respectively. CONCLUSION : The pharmacokinetics process of metformin hydrochloride in rats fits one-compartment model. Yuquan Pill has significant effect on the pharmacokinetics of metformin hydrochloride in rats.
