摘要
用麦草、蔗渣和麦麸等农业废弃物作为主要原料生产木聚糖酶,并对黑曲霉FSY01液体发酵产木聚糖酶的条件进行了优化。研究表明最适产酶条件:以麦草/麦麸3:1(w/w)作碳源,用量3%;尿素/硝酸铵1:1(w/w)作氮源,C/N比5:1;调节初始pH值4.2;培养温度30℃;培养时间72 h,在此条件下黑曲霉液体发酵产木聚糖酶最高活力达到119.6 IU/mL。添加少量可溶性低聚木糖可以提高产酶活力,加入Tween80则抑制木聚糖酶的产生。
Agricultural wastes, such as wheat straw, bagasse and wheat bran, were used as the main raw material for the production of xylanase, and the conditions of xylanase production by Aspergillus niger FSY01 through liquid fermentation were optimized. The results showed that the optimum conditions of enzyme production were: Straw / wheat bran 3:1 (w / w) as carbon resource with the amount 3% ; urea/ammonium nitrate 1:1 (w/w) as nitrogen resource, C/N ratio 5:1 ; initial pH 4.2 ; culture temperature 30 ℃ and incubation time 72 h. Under such conditions, liquid fermentation AspergiUus niger xylanase enzyme activity reached the highest 119.6 IU / ml. Addition of a small amount of soluble Xylooligosaccharides could increase the production activity, while addition of Tween80 inhibited the formation of xylanase.
