摘要
目的:对比研究常氧和低氧条件下三氧化二砷(arsenic trioxide,As_2O_3)对人肺腺癌A549细胞增殖的抑制作用。方法:分别在常氧(21%O_2)和低氧(5%O_2)条件下以1、2、4μmol/L As_2O_3处理人肺腺癌A549细胞,12、24、48 h后收集细胞,以MTT法检测细胞增殖抑制率,以Annexin V-PI双染色流式细胞术检测细胞凋亡,分析两种条件下As_2O_3对细胞增殖抑制率及凋亡率的影响。结果:常氧及低氧条件下,细胞增殖抑制率、细胞早期凋亡率及总凋亡率均随As_2O_3浓度的增加及作用时间的延长而增加(均P<0.05),晚期凋亡率变化不显著。在As_2O_3相同浓度、相同处理时间,低氧条件下细胞增殖抑制率及凋亡率较常氧条件下无明显降低。结论:As_2O_3可促进细胞凋亡,显著抑制A549细胞增殖,且在低氧条件下的上述作用未见减弱。
Objective: To compare the anti-proliferation effect of arsenic trioxide on human lungadenocarcmoma cell line A549 under normoxia and hypoxia conditions. Methods: Different concentrations (1, 2, and 4 μmol/L) of srsenic trioxide were used to treat the A549 cells at normoxia(21% O2) and hypoxia(5%O2) conditions. The cells were collected at 12, 24, and 48 h later. MTT assay was used to examine the cell growth inhibitory rate and Annexin V-PI double labeling FCM was used to determine cell apoptotic rate. To compare the influences of 2 conditions on the growth inhibitory rate and apoptotic rate of A549 cells. Results: Arsenic trioxide markedly increased the cell growth inhibitory rate, forepart and total apoptotic rates in a time- and dose-dependent manner under both conditions; the late apoptotic rates were not significantly change. The cell growth inhibitory rate and forepart apoptotic rate were not decreased under hypoxia condition compared with under normoxia when the concentration and treatment time were the same. Conclusion: Arsenic trioxide can markedly inhibit the growth and promote apoptosis of A549 cells; and this effect of arsenic trioxide is not attenuated by hypoxia.
