摘要
目的研究p27mt基因对大肠癌裸鼠移植瘤的生长抑制和抗转移作用及其机制。方法建立大肠癌裸鼠移植瘤模型,采用瘤体内直接注射法,测量瘤体体积,观察p27mt对移植瘤的生长抑制作用。分离瘤体,制作单细胞悬液,流式细胞术检测细胞周期,测定细胞凋亡。免疫组化检测瘤体组织中MMP-9的表达。结果Ad-p27mt组、Ad-LacZ组和对照组移植瘤平均体积分别为(1.94±0.67)cm3、(2.75±0.83)cm3和(3.01±0.76)cm3(P<0.05);增殖率分别为(37.34±1.45)%、(53.16±3.27)%和(54.48±2.43)%(P<0.05);凋亡率分别为(19.79±3.32)%,(6.38±4.91)%和(7.25±5.20)%(P<0.01);MMP-9的表达率为20%、75%和66.7%(P<0.01)。结论人突变p27mt基因通过阻滞移植瘤的增殖,促进移植瘤细胞的凋亡来抑制移植瘤的生长,通过降低移植瘤MMP-9的表达抑制移植瘤的转移。
Objective To study the effect and mechanism of p27mt gene for the growth inhibition and antimetastasis of human colorectal carcinoma transplanted in naked mice. Methods The model of transplanted colorectal Lovo cell line of colorectal carcinoma in naked mice was set up. Ad-p27mt was directly injected into tumor. The transplanted tumor size, cell cycle , proliferation index (PI) and apoptosis rates were observed ,MMP-9 expression levels were observed also. Results The mean volume of Ad-p27mt group, Ad-LacZ group and control group was (1.94 ± 0. 67)cm^3 ,(2. 75 ± 0. 83)cm^3and(3. 01± 0. 76)cm^3 (P〈0. 05) the growth rate was (37. 34± 1.45) %, (53.16 ± 3. 27) % and(54. 48 ± 2. 43) % (P〈0. 05) ; the apoptosis rate was( 19. 79 ± 3. 32) %, (6. 38 ± 4. 91 )% and(7. 25 ± 5.20) % (P〈0. 01 ) ; The expression rate of MMP-9 was 20% ,75% and 66. 7% (P〈0. 01 ), respectively. Conclusion p27mt gene can obviously suppress the growth of transplanted colorectal carcinoma. The mechanism dues to inhibit tumor proliferation and promote apoptosis, p27mt gene can suppress metastasis of colorectal cancer in naked mice, the mechanism partially dues to decrease the expression of MMP-9.
