摘要
目的探讨生长抑制(inhibitor of growth1,ING1)基因是否在非小细胞肺癌(non-small cell lung cancer,NSCLC)中存在突变,以确定该基因在NSCLC发病过程中的作用。方法选择突变率较高的ING1外显子2,应用聚合酶链反应-单链构象多态性分析(Polymerase Chain Reaction-Single Strand Conformation Polymorphism,PCR-SSCP)检测73例NSCLC和13例非癌肺组织中ING1的突变情况。结果未发现所扩增的片段有泳动速率的改变,没有发现额外迁移带。结论在NSCLC中ING1基因突变很少见,ING1表达产物降低可能发生于转录或转录后水平。
Objective To detect the mutation of inhibitor of growth 1 (ING1) gene in non-small cell lung cancer (NSCLC). Methods Polymerase Chain Reaction -Single Strand Conformation Polymorphism (PCR-SSCP) was used to detect mutation of ING1 exon 2 in 73 cases of NSCLC and 13 cases of non-neoplash lung tissues. Results There is no mutation of ING1 gene in 73 cases of NSCLC and 13 cases of non-neoplasia lung tissues. Conclusion Mutation of INGI gene is uncommon in NSCLC and mutation is not a reason for ING1 inactivation. Down-expression of ING1 gene may happen in transcription or posttranscription.
