缺氧条件下血管紧张素Ⅱ对血管内皮细胞线粒体膜电位的影响及金钠多的保护作用研究

摘要 目的:观察缺氧及血管紧张素Ⅱ(Ang-Ⅱ)对人大动脉血管内皮细胞(VEC)的线粒体膜电位(MMP)的影响及金钠多的保护作用。方法:建立VEC缺氧损伤模型;实验分为空白对照组、单纯缺氧组、缺氧加金钠多保护组、单纯Ang-Ⅱ作用组、Ang-Ⅱ加金钠多保护组、缺氧加Ang-Ⅱ作用组、缺氧加Ang-Ⅱ加金钠多保护组共7个组。各组细胞经过Rhodamine 123的负载后,用激光共聚焦显微镜测定VEC内Rhodamine 123的荧光强度代表MMP。结果:VEC分别经过缺氧及Ang-Ⅱ作用后,MMP显著降低(P<0.01);缺氧条件下Ang-Ⅱ引起VEC的MMP变化较单纯缺氧或单纯Ang-Ⅱ作用进一步降低(P<0.01);金钠多可抑制MMP降低,但各金钠多保护组的MMP仍显著低于空白对照组(P<0.01)。结论:缺氧及Ang-Ⅱ均可引起VEC的MMP降低,金钠多明显减轻上述影响因素的损伤、提高MMP,从而发挥对VEC的保护作用。 Objective:To study the effect of hypoxia and angiotensinⅡ(AngⅡ)on mitochondria membrane potential(MMP)in human arterial vascular endothelial cells(VEC)and the protective effect of Ginaton.Methods:Hypoxia-induced injury model of VEC was established,and then cultured VEC were divided into seven groups:control group,simple hypoxia group,hypoxia+Ginaton group,simple AngⅡ group,AngⅡ+Ginaton group,hypoxia+AngⅡ group, hypoxia+AngⅡ+Ginaton group,which were loaded with Rhodamine 123.MMP was detected by laser confocal microscopy to see if there was any difference among these groups.Results:Compared with control group,MMP significantly decreased either in the hypoxia group or in the AngⅡ group(P〈0.01).This change was clearer in the hypoxia+AngⅡ group than in the other groups(P〈0.01).MMP change could be significantly lessened by Ginaton,but MMP of each Ginaton treated group was still lower than that of the control group(P〈0.01).Conclusion:Hypoxia or AngⅡ could significantly decrease MMP of VEC and Ginaton could increase the MMP.Ginaton has remarkably protective effect on VEC injured by hypoxia and AngⅡ.
