摘要
目的建立一种准确、简便、快速的高效液相色谱法用于测定肠内制剂中谷氨酰胺的含量。方法以邻苯二甲醛作柱前衍生剂,反相C18柱为固定相,以10mmol/L磷酸盐缓冲液(pH6.85):甲醇=50:50为流动相进行洗脱,在340nm紫外波长处测定谷氨酰胺的含量。结果在100~1500μmol/L内线性关系良好(r=0.9995)。回收率为87.37%~100.99%。结论该方法简便、准确,适用于肠内制剂中谷氨酰胺含量的分析。
Objective To establish a new high performance liquid chromatography (HPLC) method for determining the contents of glutamine (Gin) in enteral nutrient. Methods Gin was derived with O-phthalaldehyde and eluted with 10 mmol/L phosphate buffer ( pH 6.85) containing methanol on reverse phase C18 column. The derivative was detected at 340 nm ultraviolet light. Results The calibration curves of Gin was linear from 100 to 1 500 μmoL/L (r =0. 9995) , and the recovery ratio was 87.37%-100.99%. Conclusions This method is simple, rapid, and effective. It can be used to determine Gin contents of enteral nutrient.
