摘要
通过凝胶电泳及细胞死亡原位末端标记(insituendlabeling,ISEL)研究了大鼠全脑缺血时细胞凋亡。结果是在轻度缺血时细胞于24h出现凋亡,重度缺血时12h出现,高峰时间均在48~72h,凝胶电泳变化较ISEL异常出现晚,分别在24h及48h。重度缺血急性期以坏死为主,迟发性损害期则以细胞凋亡为主,轻度缺血在坏死表现不显著时,仍可见明显的凋亡细胞。结果表明:细胞凋亡参与脑缺血急性期及迟发性神经元损害,轻度缺血神经细胞损害以细胞凋亡为主。
e studied the apoptosis during rat ischemia by gelelectrophoresis and in situ end labeling(ISEL).The results showed that apoptotic cells were present at 24h in mild ischemia and at 12h in severe ischemia.The peak of apoptotic cell numbers was appeared at 48 72h both two groups.The occurring of abnormality of gel electrophoresis was later than that of ISEL respectively at 24h and 48h.The necrosis plays more important role than apoptosis in neurons damage in acute period of severe ischemia,but apoptosis was more important in delayed neuron damage following ischemic insult in acute and elelayed neuron damage period.In mild ischemia there were many apoptotic cells and few necrosis cells.The results suggest that apoptosis contributed to the neuron damage following ischemic insult in acute and delayed neuron damage periods.Apoptosis may play more important role in neuron damage of mild ischemia.
