摘要
目的研究以聚乳酸羟基乙酸共聚物为囊材,制备可缓释释药的阿柔比星纳米级微粒制剂。方法运用复乳溶剂挥发法,考虑多个条件对制备工艺的影响,将阿柔比星包封在聚乳酸羟基乙酸纳米级微粒中,并采用正交设计对处方进行优化。结果制备得到的阿柔比星纳米粒表面光滑,粒径大小均匀,球形致密圆整,具有较高的载药量和包封率,且累计释放率高。结论研究微球制备工艺合理,为医药工业上制备阿柔比星聚乳酸羟基乙酸纳米级微粒提供了理论依据。
Objective To prepare a biodegradable poly(lactic-co-glycolic acid)(PLGA) nanoparticles loaded with aclarubicin.Methods Nanoparticles loaded with aclarubicin were prepared with water-in-oil-in-water double emulsification solvent evaporation process,optimized utilizing orthogonal design.Results The aclarubicin-PLGA nanoparticles,were smooth,dense and spherical,and possessed of a median diameter in a relatively narrow distribution,and had high encapsulation efficiency and drug loading,Cumulative drug release of aclarubicin nanoparticles is high.Conclusion The research provided an optimal method for the industrial preparation of aclarubicin-PLGA nanoparticles.
