奇异变形杆菌的脉冲场凝胶电泳分型研究 被引量:3

摘要 [目的]了解2006年度深圳市南山区疑似食物中毒分离的奇异变形杆菌菌株的脉冲场凝胶电泳(PFGE)基因型,探讨奇异变形杆菌的分布、流行及变异情况。[方法]奇异变形杆菌单个菌落增菌后定量,制备琼脂糖菌体包埋胶块,蛋白酶K等消化处理,限制性内切酶SfiI酶切,制备琼脂糖凝胶,脉冲场凝胶电泳获取菌株指纹图谱,图谱条带进行分子量标化,用统计软件进行聚类分析。[结果]以80﹪相似度为标准,39株奇异变形杆菌被分成24个不同的PFGE基因型.同起疑似食物中毒菌株基因型相似度高,不同起疑似食物中毒菌株基因型有交叉但存在较大差异。[结论]PFGE基因分型技术在确定菌株之间的分子流行病学关系方面是一种有效、稳定的方法,疑似食物中毒样品中分离到的奇异变形杆菌PFGE基因型别呈现多样性,同起食物中毒部分菌株存在着不同程度的亲缘关系,不同起间存在差异。 [ Objective] To understand genotyping of proteus mirabilis (P. mirabilis) by pulsed-field gel electrophoresis that isolated from suspected food poisoning in Nanshan District of Shenzhen in 2006, and to investigate distribution, epidemic and variation of P. mirabilis. [Methods] Quantitatively measured P. mirabihs after single colony enrichment, embedded organisms in agarose, lysed the organisms in situ and treated by proteinase K, digested the chromosomal DNA with restriction endonucleases SfiI, Slices of agarose containing the chromosomal DNA fragments were inserted into the wells of an agarose gel, and subjected to PFGE to visualize the DNA fingerprinter, then mapped the molecular weight of the DNA fingerprinter band followed by cluster analysis. [ Results] Twenty-four different PFGE patterns were observed in the 39 P. mirabilis strains, with similarity 80% as criteria. Strains with same suspected food poisoning showed a high genotype similarity, strains from different suspected food poisoning produced cross-genotype but significant difference. [Conclusion] PFGE genotyping is an effective, stable way for identifying strains in the molecular epidemiology of relations. There is PFGE genotypes diversity among P. mirabilis strains from suspected food poisoning samples, isolates from same food poisoning have varying degrees of genetic relationship in contrast differences among isolates from different food poisoning.
