柠檬黄对泥鳅的急性毒性及遗传毒性实验 被引量:16

摘要 [目的]研究柠檬黄对泥鳅红细胞细胞核的毒性影响。[方法]以柠檬黄作为诱变剂,通过急性毒性实验研究了柠檬黄在不同浓度(7.482 08、.017 08、.590 09、.204 0和9.862 0 g/L)和不同染毒时间(81、62、4、324、0 h)条件下对泥鳅红细胞的损伤情况。[结果]柠檬黄对泥鳅的24 h半致死浓度LC50为8.594 0 g/L和48 hLC50为7.173 0 g/L,其安全浓度为1.515 0 g/L。柠檬黄在较低的浓度下(<0.094 7g/L)无明显的遗传毒性效应。达到一定浓度(>0.378 8 g/L)之后所诱导的微核率基本上随着浓度的加大和染毒时间的延长而增高,达到最高值后,开始下降。柠檬黄诱发的微核率与对照组相比达到显著或极显著差异水平。[结论]柠檬黄达到一定的浓度和染毒时间之后对泥鳅具有一定的遗传毒性。建议食品加工业在使用柠檬黄时,应严格限量使用。 [Objective] The research aimed to study the toxicity effects of tartrazine on the erythrocyte nucleus of Paramisgurus dabryanus.[Method] In the acute toxicity experiment with tartrazine as the mutagen,the damage of tartratine to the red blood cell of P.dabryanus under the condition of different tartrazine concentrations(7.482 0,8.017 0,8.590 0,9.204 0 and 9.862 0 g/L resp.)and different contamination times(8,16,24,32,40 h resp.) was studied.[Result] The median lethal concn.(LC50) of tartratine to P.dabryanus at 24 h and 48 h were 8.594 0 g/L and 7.173 0 g/L respectively,and its safe concentration was 1.515 0 g/L.When tartrazine was at a lower concentration(〈0.094 7 g/L),no obvious genetic toxicity effects existed.When the concn.of tartrazine arrived at a certain level(〉0.378 8 g/L),the micronucleus rate induced by tartrazine was basically increased with the concn.of tartrazine increasing and delay of contamination time.After achieving the maximum,the micronucleus rate started to drop.The micronucleus rate induced by tartrazine arrived at significant or extremely significant difference level compared with the check.[Conclusion] When the concentration of tartrazine arrived at a certain level and some contamination time,the tartrazine had certain genetic toxicity effects on P.dabryanus.It is suggested that the amount of tartrazine used in food processing industry should be severely limited.
