摘要
目的观察蝎毒抗癌多肽对卵巢癌细胞HO-8910生长的抑制作用。方法卵巢癌细胞HO-8910分为空白对照组和蝎毒抗癌多肽处理组(终浓度分别为1、100、200、400、800 mg/L),采用细胞毒试验(MTT)法检测蝎毒抗癌多肽对卵巢癌细胞HO-8910生长抑制率,流式细胞术检测细胞周期和细胞凋亡的变化。结果1、100、200、400和800 mg/L组HO-8910细胞生长抑制率分别为2.56%、12.47%、28.53%、47.93%和66.92%,200、400和800 mg/L组与对照组比较差异具有显著性(P<0.01);在400和800 mg/L组,与空白对照组比较,400 mg/L组HO-8910细胞S期细胞数明显减少(P<0.01),G2+M期细胞明显增多(P<0.01),800 mg/L组HO-8910细胞G2+M期细胞数明显减少(P<0.01),G1期细胞数有增多趋势;与空白对照组比较,400和800mg/L组HO-8910凋亡细胞数明显增加(P<0.01)。结论在一定范围内蝎毒抗癌多肽可明显抑制卵巢癌细胞HO-8910的生长,其机制与蝎毒抗癌多肽抑制HO-8910细胞DNA合成、诱导HO-8910细胞发生G2+和G1期阻滞及诱导HO-8910细胞凋亡有关。
Objective To explore the inhibitory effects of polypeptide from buthus martensii venom on the growth of HO-8910 ovarian carcinoma cells. Methods HO-8910 ovarian carcinoma cells were divided into two groups: control group and treatment group with polypeptide from buthus martensii venom (final concentration as 1, 100, 200, 400 and 800 mg/L, respectively) . MTT method was used to detect the inhibitory rate of tumor cells treated with polypeptide from buthus martensii venom in vitro and flow eytometry was further adopted to observe the changes of cell cycle and apoptosis of HO-8910 ceils. Resuits Polypeptide from buthus martensii venom could inhibit the growth of HO-8910 ovarian carcinoma cells significantly at the concentrations of 200, 400 and 800 mg/L and the inhibitory rates were 28.53% , 47.93% and 66. 92% , respectively (P 〈 0.01 ) . Compared with control, cells in S stage were decreased and cells in G2 + M stage were increased obviously (P 〈0. 01 ) after polypeptide from buthus martensii venom was given at 400 mg/L, while cells in G2 + M stage were decreased (P 〈0. 01 ) and cells in G1 stage had a ascending thread after treated with 800 mg/L polypeptide from buthus martensii venom. Furthermore, apoptosis could be induced obviously by polypeptide from buthus martensii venom at 400 and 800 mg/L compared with control ( P 〈 0. 01 ) . Conclusion Polypeptide from buthus martensii venom could inhibit the proliferation and growth of HO-8910 ovarian carcinoma cells and the chages of cell cycle and apoptosis may be important mechanism of inhibitory effects of polypeptide from buthus martensii venom.
