摘要
目的探讨增殖细胞核抗原(PCNA)反义寡核苷酸对裸鼠骨肉瘤生长的抑制作用。方法制备BALC/C裸鼠皮下骨肉瘤模型12只,随机分为PCNA反义寡核苷酸治疗组和对照组,每组6只。接种MG-63人成骨肉瘤细胞后24 h内用PCNA反义寡核苷酸皮下注射进行治疗,对照组只注射生理盐水,每周2次,连续4周;观察各组裸鼠肿瘤的生长情况、裸鼠的一般情况与生存期。断颈处死裸鼠,游标卡尺测量肿瘤体积大小,天平称量肿瘤的重量。结果PCNA-ASODN治疗组裸鼠骨肉瘤的生长受到抑制,抑瘤率为46.53%。结论PCNA反义寡核苷酸对骨肉瘤的生长具有抑制作用,通过反义技术可抑制靶基因的表达,从而抑制骨肉瘤细胞的增殖。
Objective To study the inhibitory effect of PCNA antisense oligonucleotides on the growth of osteosarcoma in nude mice. Methods The osteosarcoma cells were cultured and implanted subcutaneously into 12 nude mice which were subsequently divided into two groups: PCNA - ASODN group and the control group. The treatment was given PCNA - ASODN at 24 hours after cell inoculation. The weight and volume of subcutaneous tumors were measured. Result The tumor groxah of PCNA - ASODN group 3.98 ± 0.78 g was significantly inhibited. The inhibition rate of tumor growth of the PCNA - ASODN group was 46.53 %. Conclusion It was demonstrated that the groxah of osteosarcoma in nude mice can be inhibited by PCNA antisense oligonucleotides. The expression of PCNA target gene and prolification of osteosarcoma cells can be inhibited by PCNA antisense oligonucleotides.
