摘要
目的:研究蒙药制剂"三花散"对小鼠免疫水平的影响,进一步探讨蒙药制剂"三花散"的免疫药理机制。方法:常规方法制备蒙药"三花散"醇提液为实验药物,以小鼠为试验动物,通过灌胃法连续给药,观察不同剂量的蒙药制剂"三花散"分别对小鼠淋巴细胞转化率、腹腔巨噬细胞吞噬功能影响,RT-PCR技术检测对免疫分子Fas配体及穿孔素等表达水平的影响。结果:实验结果表明,蒙药制剂"三花散":(1)能够提高小鼠腹腔巨噬细胞吞噬率和吞噬指数,与正常对照组比较差异显著(P<0.01);(2)能够促进小鼠体内淋巴细胞的转化与增殖,实验组小鼠的淋巴细胞转化率较正常对照组显著提高(P<0.01);(3)实验组小鼠脾脏免疫分子Fas配体及穿孔素为阳性表达,实验组与对照组比较差异显著(P<0.01)。结论:蒙药制剂"三花散"不仅能使小鼠腹腔巨噬细胞吞噬功能及淋巴细胞活性增强,并能增强小鼠脾组织免疫分子Fas配体及穿孔素的表达水平,提示蒙药制剂"三花散"能够有效的促进小鼠的免疫功能。
Objective:The purpose of the test was to find the effects of Mongolia Traditional Medicine Preparation Sanhuasan (SHS) on immune level in mice and probe into the effects of SHS on immune mechanism system so as to offer scientific basis for clinic application of Mongolia Traditional Medicine Preparation SHS, realize the modernization of national medicine and make contributions to the cause of human health. Methods : This experiment used SHS as the experimental medicine, and mice as the experimental animals. It choosed pure Balb/c mice, Mongolia Traditional Medicine Prep- aration SHS of different doses were fed to mice continuously in force . After several days ,the samples in each group were collected for testing. It demonstrated through the transformation mice of lymphocytes, the phagocytes percentage and index of peritoneal macrophage that the effect of Mongolia Traditional Medicine Preparation SHS on the mice. Moreover,the two indexes about the lymphocytes activation by RT- PCR, such as perforin and fasl were observed. Results: The results showed as follows : group I and 2 could significantly increase the transformation mice of lymphocytes compared to the contrast group ( P 〈 0. 01 ) ; it showed that SHS could significantly increase cellular immune function of mice; SHS could significantly enhance the phagocytic activity of peritoneal macrophage compared to the contrast group ( P 〈 0. 01 ) ; it also showed that SHS could promote the nonspecific immune function of mice ; These were enhanced significantly when the immunology function of mice was inhibited by Cy ( P 〈 0.01 ) ;it showed that the SHS enhanced the killing activity of the lymphocytes obviously - exhibiting as the expression of the two ( Fasl and perforin) index increasing obviously. Conclusion:These experiments indicated that Mongolia Traditional Medicine Preparation SHS was able to activate immune function of mice and enhance Fasl and perforin to express in splenetic. These tests will provide sufficient scientific basis for clinic application of Mongolia Traditional Medicine Preparation SHS.
