摘要
目的:研究银杏叶提取物抗脑缺血再灌注损伤作用与线粒体ATP敏感性钾通道(mitoKATP)开放的关系。方法:用大脑中动脉线栓法(MCAO)制作大鼠局灶性脑缺血再灌注损伤模型。造模前静脉给予银杏叶提取物15mg·kg-1和(或)选择性mitoKATP拮抗剂5-羟基癸酸(5-HD)10mg·kg-1预处理,以脑梗死体积、神经缺陷评分、SOD活性和丙二醛含量,评价银杏叶提取物抗脑缺血再灌注伤的作用与mitoKATP的关系。结果:银杏叶提取物具有改善大鼠缺血再灌注引起的脑功能和组织损伤,但此作用可被预先给予的5-HD减弱。结论:银杏叶提取物抗脑缺血再灌注伤作用与mitoKATP开放有关。
Objective :To study on the relationship between the protection of ginkgo biloba extract( Egb761 ) against focal cerebral ischemia- reperfusion injury and mitochondrial ATP- sensitive potassium channels(mitoKATP) in vivo. Methods:A reversible middle cerebral artery occlusion model(MCAO) was used in this study. Fourty male Sprague -dawley rats were divided into four groups (n = 10) : MCAO group, Egb761 group ( 15mg·kg^-1), 5 - HD group ( 10mg·kg^-1) ,5 -HD + Egb761 group. The neurological funciton was scored and the infarct volume,activity of SOD,content of MDA were measured after operation. Results :In 5 - HD + Egb761 group, the infarct volume, the neurological funeiton were significantly higher than those in Egb761 pretreated group. Conclusion :The result is suggested that the protects of Egb761 against focal cerebral isehenfia - repefusion injury may be related to mitoKATP.
